with (4) mixed connective tissue disease (MCTD) and (15) systemic lupus
erythematosus (SLE) were studied. Two SLE sera reacted with U1RNP and not
with Sm. Immunotblots (IB) were performed on human spleen (HS) for the detec-
tion of Ro and La and on rabbit thymus (RT) extracts for Sm and U1RNP.

Dissociation of Sm and RNP reactivity was best associated with the 68 kD prote-
in. RNAse treatment of the extract removed the 68 and revealed the 44/42/38 kD
proteins. Standard Ro antiserum reacted with both 57 and 45 kD proteins. Anti-
La antibodies bound a triplet of 30/28/27 kD proteins on HS extracts treated by
trypsin. In addition to the good correlation with CIE, IB on tissue extracts reveal-
ed the protein specificities associated with each antigenicity.

Analysis of the 60 kD Ro Protein - hY RNA Complex: An RNA

Ro (SS-A) antigen is a common autoimmune specificity and exists in humans
as an inabundant ribonucleoprotein complex (RNP) composed of a 60 kD protein
and one of a series of small RNA polymerase III products, designated hYl-hY5
RNA. Detailed information on the structure and function of the Ro RNP is lack-
ing. We have recently reported the isolation and characterization of cDNA clones
encoding the 60 kD human Ro protein (DEUTSCHER et al. PNAS 85,
9479-9483). The Ro amino acid sequence contains in region of similarity to the
