Autoantibodies to Nonhistone Chromosomal Proteins HMG-1 and
HMG-2 in Subsets of Juvenile Chronic Arthritis (JCA)

Antihistone antibodies (AHAs) in sera of patients suffering from juvenile
chronic arthritis (JCA) have been reported recently by several investigators [1, 2].
In our study of 100 patients with JCA admitted to the Rheumatic Children’s
Hospital in Garmisch-Partenkirchen we observed, in addition to antibodies
against various histones, autoantibodies against the nucleosomal proteins
HMG-1 and HMG-2. All sera tested were ANA positive by indirect immunofluo-
rescence on HEp-2 cells and negative when tested against double stranded DNA
by an indirect solid phase ELISA (Progen GmbH, Heidelberg). In the analysis re-
ported here we submitted purified HMG-1 and HMG-2 from calf-thymus [3] to
PAGE and subsequent western blots. The results presented in the table give a cor-
relation between clinical diagnosis and the presence of autoantibodies to
nucleosomal proteins HMG-1 and HMG-2 in JCA patients.

Grouping the sera tested according to the clinical diagnosis (Table 1) reveals
a correlation between detectable amounts of antibodies to the chromosomal pro-
teins HMG-1 and HMG-2 and subsets of JCA. JCA patients with polyarthritis
react to 28%, whereas patients with oligoarthritis show a higher incident of 43%
binding to the high mobility group proteins 1 and 2.
